The Pоlymerаse Chаir Reаctiоn (PCR) has revоlutionized molecular biology by allowing biologists to amplify and alter the sequence of DNA in situ. A typical PCR reaction repeatedly cycles over a three step change in temperature to facilitate different parts of the reaction.  Researchers provide template DNA at the start of the reaction while each cycle generates new strands of template, allowing exponential amplification of target DNA.  In the first step a PCR reaction, the double stranded DNA template which will be amplified must be separated into single strands at [blank1], called the [blank2] step.  The second step of any PCR reaction requires hybridization of short DNA strands, called primers, to the single-stranded DNA template.
