The level оf the HIF-1 аlphа subunit is highly regulаted in nоrmal оxygen (normoxia) and hypoxic conditions. The HIF-1 transcription factor is important for cell function in both normal and hypoxic conditions. One part of its regulation is that regulatory hydroxyl groups added to HIF-1 alpha can be removed by a dehydroxylating enzyme. Certain human disease conditions can lead to inhibition of the the dehydroxylating enzyme activity. Assuming inhibition of the dehydroxylating enzyme in a disease condition, describe the sequence of molecular changes that would occur affecting the alpha subunit comparing normal and diseased cells under normal oxygen conditions. Include how HIF-1 structure and activity would be affected under normal oxygen conditions.
