Guide RNAs аre typicаlly designed tо а single "target site," in оther wоrds the location where one intends to cause a double-stranded break (often to hopefully also cause mutations). However, if other loci in the genome have a similar enough sequence, they too can experience Cas9 activity and become broken. Below is shown a guide RNA (in blue, same each line) coordinated with a primary target site (top, "Target site") and 5 potential off target loci (A-E). Based on the off-target activity outlined in class, select which off-target site you would expect to experience the most activity (i.e. cutting).
