Cоnsider the Sоnic hedgehоg (SHH) signаling pаthwаy depicted below. A description of the pathway is also included.  QUESTION: What would happen to the cell if a mutation occurred in the gene coding for Patched, causing Patched to lose its ability to bind SHH? LEFT: Diagram shows what is happening in the cell when SHH is NOT present. The receptor Patched inhibits the protein Smoothened in the cell. When Smoothened is repressed, Gli proteins are tethered/tied to microtubules in the primary cilium by proteins Sufu and Cos2. Because Gli is tethered/tied there, the protein PKA can turn Gli into its repressor form by cleaving it. This Repressor form of Gli travels into the nucleus and inhibits the transcription of Hedgehog-responsive genes.  RIGHT: Diagram shows what is happening in the cell when SHH IS present. SHH binds to its receptor Patched, and this stops Patched from inhibiting the protein Smoothened. Smoothened is then able to travel into primary cilium, and inhibit the proteins Sufu and Cos2 from tethering/tying Gli to the microtubule. This allows Gli to avoid being cleaved by PKA and to therefore transform into its activator form. This activator form of Gli travels into the nucleus and activates transcription of Hedgehog-responsive genes.
