You use your ligation reaction to transform E. coli Z-competent cells, plate them on LB medium containing IPTG (to provide access to the lac promoter), X-Gal (a colorless substrate for beta-galactosidase) and an antibiotic, and incubate them overnight at 37C. The next morning you see lots of colonies, both white and blue.
