This twо pаrt questiоn аddresses the expected results оf experiments using C57Bl/6 mice (genotype H-2 b/b). These experiments аre based on the study by Zinkernagel, as discussed in class, that used a chromium release assay to measure the killing activity of splenic CTL (effector cells, see figure). Virus-infected fibroblasts (target cells) are incubated with 51Cr which is taken up into the cytosol of each target cell. After washing, the target cells are co-incubated with spleen cells. If any of the target cells are killed, 51Cr is released, and the amount of radioactivity in culture supernatant will be proportional to the number of killed cells. Part 1: C57Bl/6 mice are infected with virus X. Two weeks later, the killing activity of their spleen cells is measured using a chromium release assay and target cells that were also infected with virus X.  To do so, you set up three different cultures using target cells having different H-2 genotypes as follows: b/b, d/d, b/d. Question 1: Which of these target cells will be killed by the C57Bl/6 killer cells? (4 pts) Part 2: Before infecting the mice, you lethally irradiate them to destroy all immune cells, and then you inject them with bone marrow taken from other mice so they can remake their immune systems.
