The nature of the NMR pulse sequence is such that: (a) the “intra” signals typically have significantly higher intensity than the “inter” signals and (b) Ca and Cb signals have opposite phase. The CBCA(CO)NH is acquired to eliminate any ambiguities between “intra” and “inter” signals, as it correlates the amide 1H-15N chemical shifts of one amino acid to the Ca and Cb chemical shifts of only the previous (inter) amino acid. In the latter case, both the Ca and Cb signals have the same phase.
