During DNA synthesis, there are a number of components (enzymes, nucleic acid sequences, nucleotides, binding proteins, etc.) necessary to separate DNA strands, prevent them from reconnecting, starting, and continuing synthesis of new, complementary strands of DNA. Figure 1 is adopted from your textbook and shows some of the steps in this process for the leading DNA strand. Polymerase Chain Reaction (PCR) is an in vitro (not inside cells) method that allows researchers to synthesize specific sequences of DNA in the lab. The process of PCR consists of several steps of heating and cooling a tube containing DNA: heating it to approximately 95°C to separate the strands, cooling it to encourage primer sequences to attach to the DNA, and then heating it to approximately 70°C for the polymerase to build the new DNA strands. This heating and cooling is repeated about 30 times, each time doubling the amount of DNA sequences that the researchers are targeting.
