Upоn binding tо аn extrаcellulаr signal, a pair оf RTKs dimerize and phosphorylate three tyrosine residues on each other’s cytoplasmic tails. These tyrosines serve as docking sites for three intracellular signaling proteins: phosphoinositide 3-kinase (PI3K), protein kinase X (PKX), and an adaptor protein (adaptor 1). To determine which tyrosine is recognized by which downstream signaling protein, investigators construct a series of mutant receptor proteins in which the tyrosine residues are replaced, one at a time, with an alanine residue, preventing phosphorylation at that site. The proteins bound to each of these mutants are experimentally identified (by co-immunoprecipitation) and the results are shown in the table.
