IL6-3 In а 2011 pаper, Cussigh et аl examine the assоciatiоn between prоmoter polymorphisms in the interleukin 6 (IL-6) gene and chronic hepatitis C virus (HCV) infections to determine if these polymorphisms were a) influencing viral clearance (can the host get rid of the virus) and/or b) influencing disease progression. Chronic infection can result in a number of outcomes ranging from minimal liver damage, to cirrhosis, to hepatocellular carcinoma. They used a case-control study design, and the initial results are shown in Table 2 below. The authors go on to show a relationship between promoter polymorphisms and disease progression in chronic HCV patients. They identified three phenotypic groups – high IL-6 producers (HP, high circulating level of IL-6 protein), intermediate IL-6 producers and low IL-6 producers (IP and LP). They examined the proportion of individuals of each phenotypic class in controls (CS), and in HCV patients with severe (bad) outcomes (CH) and HCV patients with less severe outcomes (CL) (Figure 2 below). A diplotype is just a haplotype that contains two variable sites, in this case sites -597 and -174 indicated in Table 2 above.
