(must be specific to your tissue type—for example biocompatible and good mass transport are not specific and is therefore not a correct response here) Choose specific materials and state why you would use the chosen materials for this tissue type. (decellularized tissues are not allowed) Does this material degrade? By what method? (be specific with type of degradation and erosion mechanism) If not, how will you modify it so it does degrade, and how does the modified material degrade? How can you tune degradation to match regeneration of tissue? (be specific—how to slow it down, how to speed it up and what relative rate do you need?) Choose a specific method of scaffold fabrication and state why this method synergizes with the needs/functionality of your chosen tissue. State one specific biomolecule (growth factors, cytokines, or hormones that are normally found in the body with this tissue type, and is NOT an ECM protein) that promotes your tissue’s successful regeneration, state why it is important, and state how you will incorporate it into your scaffold. (just stating “added to scaffold” is not enough—you must state specific techniques. Select from drug loading techniques and/or bioconjugation techniques learned in lecture) Choose one scaffold design parameter that can be modified to promote your tissue’s successful regeneration, state why it does, and state how you will modify this parameter (i.e., pore size, surface chemistry, topography, etc.). Choose a specific cell type to use with or within your scaffold, state the source of the cell, and why you are choosing it. If you choose to start with stem cells, make sure you specify how you plan to differentiate them into the appropriate cell type, and state what cell type/s you need for your tissue type. Choose a specific bioreactor to encourage successful development of your scaffold and state why the selected tissue and scaffold design benefit from this bioreactor.
